We are excited to share our recently published interdisciplinary paper that bridges organic chemistry, analytical chemistry, bone physiology, and bioinformatics to explore the lipogenic mechanisms of thiazolidinediones (TZDs) on osteogenesis and adipogenesis. Our research reveals that the new generation of TZDs enhances peripheral insulin sensitivity while preserving bone structure.
The accompanying image illustrates the divergent fates of bone marrow mesenchymal stem cells when exposed to TZDs, leading to either fragile or healthy bone. Our study demonstrates that inhibition or silencing of the mitochondrial pyruvate carrier plays a crucial role in this process, as evidenced by 13C metabolic flux analysis.
Key takeaways from our research:
1. Subcellular fluxomics: We developed a novel approach to infer metabolic fluxes in mitochondria and cytosol under physiological conditions. This method combines bioorthogonal click chemistry, LC-MS/MS, and 13C-labeled tracers, providing unprecedented insights into compartment-specific metabolism.
2. Bioorthogonal mitochondrial probes: We designed innovative probes targeting mitochondrial carbonyl-containing metabolites using a modified TPP+ moiety. This advancement allows for precise tracking of mitochondrial-specific metabolic processes.
3. Complex lipid metabolic flux analysis: By employing isotope labeling techniques, we conducted simultaneous analysis of multiple lipid pathways. This comprehensive approach yielded quantitative insights into lipid flux distributions, enhancing our understanding of lipid metabolism dynamics.
