Is #AI hitting a plateau in structure prediction? Help us find out at CASP17! 🧪🧬 Calling for Targets: Immune Complexes, protein - ligand complexes, RNA/DNA, conformational ensembles, membrane proteins, viral origins, and large complexes. The Rule of Thumb: If AF3 can’t model it, we want it.

The Critical Assessment of Structure Prediction (CASP) experiment is calling for prediction targets: Immune Complexes, Organic Ligand-Protein Complexes, Nucleic Acids and Complexes, Conformational Ensembles, Difficult Protein Structures and Complexes. 
Rule of Thumb: If AlphaFold3 can generate a high-quality model, it is likely not a CASP-grade challenge. If it struggles, we want it.

The beauty of symmetric, homo-oligomeric biomolecular complexes cannot be denied— nature builds these from RNA as well! Here’s to the years ahead, as the community uncovers what these assemblies can teach us about RNA biophys and functions, and how they may inspire future biotechnology.

OLE RNA from Clostridium botulinum (pdb_00009lcr) is shown in pink on the left. ROOL RNAs can form cages built from 6 (right, top, pdb_00009j6y) or 8 (right, bottom, pdb_00009mds) subunits.

When I determed OLE's structure, a transmembrane RNP was staring at me. Took a little bravery to propose this in our manuscript, so I am stoked to see the experts elaborate further. Past data does not rule out OLE RNA spanning the membrane, but data is still needed. Excited for future OLE studies!

What is the status of nucleic acid structure prediction? Our analysis of CASP16 (doi.org/10.1101/2025...) reveals human expertise is still necessary for the most accurate prediction, but accuracy still heavily relies on templates; having seen a similar structure already.
