The +1 nucleosome functions in RNA Pol II transcription initiation and the transition to elongation

By combining biochemical assays and electron cryo-microscopy, Zhan, Abril-Garrido, et al. elucidated key steps of transcription initiation and the transition to elongation in the presence of the +1 nucleosome. The authors demonstrated for the first time that the +1 nucleosome has an active role in the initiation-elongation transition.

RNA reinforces condensate nucleation on chromatin to amplify oncogenic transcription

Budinich et al. show that locally produced transcripts reinforce the nucleation of pathogenic condensates formed by mutant ENL at permissive target loci. This RNA-enhanced condensate nucleation in turn increases mutant ENL occupancy and transcriptional bursting at key oncogenic targets, thus surpassing the gene activation threshold required for tumorigenesis.
