Anybody be able to help me! Journal is being picky, I've uploaded the sequences to NCBI for a publication and selected for them to be published in a year or on publication but the journal are insisting that they won't review unless they are public now. How can I change this? #microsky

It's official I'm giving up. Transformation of a full 5.6 kb operon was always a tough ask but these poorly E. coli really are rejecting the operon 😢 1 week to achieve OD0.1!

Can we make working individually normal again. Collaboration is great don't get me wrong, but as a neurodivergent individual communication is the bit of science I'm worst at. I can design awesome programs of work, dream up experiments, write grants (and papers if I have a fixed deadline).
Is there anything more frustrating than reading "we did the protocol of Z with the following modifications" only to find Z saying "we did the protocol of Y with the following modifications" only to find Y saying "we did the protocol of X with the following modifications" 🙄

The biggest tragedy with science is the loss of materials. Someone publishes something and the assumption is that it can be made available for future use. This falls down when a group leader retires. A combination of their group moving on and loss of contacts.
Alas my ERC starting grant was not passed to stage 2. I always knew my goal was just to get the second part read and this just feels like one more kick in the teeth. Guess I'll buy some new jumping spikes tonight....

I recently sequenced an interesting bacterial genome, with the idea of making the interesting recombinant genes from this strain. Problem: after a morning searching the genes of interest just aren't there. Not a trace, and all literature says there should be 2 copies. 😭🤯 #help
