Google replaced pushing Git tags for certain source code with obtaining source code via Google Drive after making a request through Google Forms. It's completely ridiculous and they've gradually become very slow at handling requests. They're in clear violation of the GPLv2 now.

My take on p-values: 1. p-values do what they are designed to do, which is not what most people using them imagine them to do. 2. Trying to fix p-values is a trap, because no single-number summary can do what people want p-values to do! 3. Trying to explain p-values is also a trap, because...

This is a real image. Taken by Voyager 1 in 1979, the icy moon Europa floats above Jupiter's swirling Red Spot maelstrom. The shadow is from Io, another of Jupiter's large inner moons, which is out of frame. Oh to see this view with my own eyes (safely shielded from the lethal radiation fields)

The swirling face of Jupiter, with the Great Red Spot at lower right. At lower left, difficult to discern from the cloudy backdrop, is Europa, lit from the right. At upper right is a dark circle—the shadow of Io.

Exciting news 📣 The first preprint from @grandlab.bsky.social is out 🧬 How are essential genes controlled? By rapid degradation and recovery of TFs alone or in combination, we show that essential genes rely on a single dominant TF, despite dense co-binding. www.biorxiv.org/content/10.6...

Essential genes are dominantly activated by single transcription factors

Cell viability depends on the precise expression of essential genes, which are controlled by CpG-island (CGI) promoters densely bound by transcription factors (TFs). This has led to the prevailing model that TFs cooperate to ensure ubiquitous expression. Here, using rapid and reversible single and combinatorial degradation in murine stem cells, we systematically dissect the regulatory interactions between five key TFs. We uncover an unexpectedly specific architecture in which regulatory dominance, rather than cooperation, is the prevailing mode, where individual TFs autonomously drive chromatin opening and gene activation at largely distinct promoters. Cooperative regulation occurs at a minority of sites with antagonistic or synergistic outcomes modulated by the interplay between nucleosome positioning and TF sensitivity to chromatin. This logic is recapitulated at synthetic sequences and reflected in human genetic variation. These findings reveal that single TFs dominantly activate distinct sets of CGI-linked genes, including essential genes, across development, homeostasis, and disease. ### Competing Interest Statement The authors have declared no competing interest. Just found out about the new science journal "Stacks". The editorial board includes the legendary @elisabethbik.bsky.social. Publication costs are what they should be. Moreover, it has an interesting peer-review where reviewers interact, agree and vote for publications. stacksjournal.org

Stacks Journal | Create better science, together.

A scientific journal designed for ease and ethics. Publishing open-access articles, Special Issues, and entire journals.

Oligarchs have always been a threat, but tech oligarchs represent a new level of danger. Their wealth dwarfs historical tycoons and their control over algorithms allows them to curate human reality. They own the digital infrastructure of thought
