Challen and colleagues show that the clonal expansion of Dnmt3a-null HSCs in serial bone marrow transplantation assays is not dependent on the loss of DNA methylation functions of Dnmt3a. They identify non-canonical functions for Dnmt3a in the regulation of telomere length and genome stability that restrict the functional potential of HSCs.

Xing et al. construct a comprehensive pan-cancer human brain metastasis (BrM) atlas at single-cell resolution, uncovering five major BrM ecotypes associated with distinct phenotypic characteristics. The study also identifies several novel cell states and subsets that are strongly enriched in BrM across different lineages, suggesting them as potential therapeutic targets for future investigations.
