Exciting preprint from @hessofarhan.bsky.social! Mechanical stress has a new sensor! IRE1 detects ER membrane tension and drives protein synthesis independently of its canonical UPR role, reshaping cell and organelle mechanobiology. www.researchsquare.com/article/rs-1...
Mechanosensing at the endoplasmic reticulum by IRE1
Sensing and integration of mechanical forces in eukaryotic cells have largely been attributed to the plasma membrane and the nucleus. Here, we identify the endoplasmic reticulum (ER) as an autonomous ...

Yeast cells utilize a TORC2 feedback loop for plasma membrane adaptation to mechanical stress, with increased sterol transport from plasma membrane to endoplasmic reticulum activating TORC2 signaling Robbie Loewith, Aurélien Roux @rouxlab.bsky.social and coworkers www.embopress.org/doi/full/10....
🚨 Lipid Time! 🚨 Excited to share our latest story now on @biorxiv-biochem.bsky.social ! Our brilliant PhD student @manpreet0700.bsky.social , together with @walter-nickel-bzh.bsky.social and myself, shows how PI(4,5)P₂ asymmetry in the plasma membrane drives the unconventional secretion of FGF2! 👇👇👇
Plasma membrane transbilayer asymmetry of PI(4,5)P2 drives unconventional secretion of Fibroblast Growth Factor 2
Unconventional secretion of Fibroblast Growth Factor 2 (FGF2) is based upon direct self-translocation across the plasma membrane, a process that involves the transient formation of a lipidic membrane ...

This yielded beautiful tomograms that showed that microvillar actin is first reabsorbed from the microvillar base, gets repositioned underneath the plasma membrane of the cell body, and is then remodeled from a set of bundles into a meshwork, with the typical architecture of a cortex.
Previous preprint is now published in RSC Chemical Biology. If you are interested in plasma membrane labeling, see the paper! 👇 pubs.rsc.org/en/content/a...
Plasma membrane labelling efficiency, internalization and partitioning of functionalized fluorescent lipids as a function of lipid structure
Labeling the plasma membrane for advanced imaging remains a significant challenge. For time-lapse live cell imaging, probe internalization and photobleaching are major limitations affecting most membr...

New paper out! Would you like to fluorescently label the plasma membrane in live, fixed or permeabilized cells? I tested different lipid structures and labelling approaches to decide what makes a good membrane labelling probe for such imaging experiments! www.biorxiv.org/content/10.1...
New: Using F-tractin to visualize #actin in living cells? Dmitry Shatskiy, Athul Sivan, Roland Wedlich-Söldner, and Alexander Belyy @alexanderbelyy.bsky.social determined the F-tractin-F-actin structure and showed that this probe is nearly identical to #Lifeact. https://buff.ly/3EIqAur #cytoskeleton
It's been a while in the making, but finally out:😂🥳 We’re excited to share our new work with H. Pavenstädt, D. Braun, M. Krahn, P. Bieling and @raunser-lab.bsky.social on INF2 and global actin remodeling! Read the story here: www.biorxiv.org/content/10.6... #Actin #Cytoskeleton #CellBiology #INF2

The Myo2 adaptor Ldm1 and its receptor Ldo16 mediate actin-dependent lipid droplet motility
Zhao et al. identify the molecular machinery for lipid droplet motility, consisting of the type V myosin Myo2, the Myo2 adaptor protein Ldm1, and its surface receptor Ldo16. They uncover a molecular l...

We start the second day of the #BiomembraneDays25 with impressive results on phase separation in the yeast plasma membrane by Roland Wedlich-Söldner
Our study explains why yeast cells lacking Neo1 are neomycin sensitive, which targets PI4P when it is exposed on the outer leaflet of the plasma membrane. We show that Neo1 flips PI4P, offering key mechanistic insight into how phosphoinositide membrane asymmetry is maintained. @vuartsci.bsky.social
Very excited to announce the #DGZ awardees 2025. We look forward to welcoming all award winners to our meeting in Heidelberg, where they will present their work!
