We map out the carbon fixation pathway for the first time in a multicellular alga, the green seaweed Ulva, and reveal a unique Rubisco-Linker interaction in the pyrenoid, a chloroplast condensate. Some progression on 58 kDa GBA as part of my work with @recon4imd.bsky.social. Combined tilted data (Glacios) with careful processing gives a ~4.0 Å map with clear secondary structure features. Still not the resolution that I need for variant studies but a step in the right direction! #CryoEM

Other than the practical value (and its or huge value!) this final LEA experiment really shows how complex the air water interface is in cryoEM

5/5: Combining datasets collected with different LEA proteins computationally produced more isotropic reconstructions. Surprisingly, premixing multiple LEA proteins before grid preparation did not simply combine their effects and produced unexpected orientation distributions.

someone in the lab needs to use the PrepCell and apparently i am the last person to have used it 🙃. Needless to say there is a lot of sceptisism. But i have the receipts! Had to explain about degassing polyacrylamide, and analog recorders! lets see whether we can get this puppy to work.
