This Is What Drives Axon Growth. Axon growth is powered by the actin cytoskeleton at the growth cone. Here, filamentous actin dynamics drive fast, directed axon extension in real time. #Fluorescencefriday #science #neuroscience #microscopy 🧪
Proximity labeling users: check out this study by @zhixingchen2.bsky.social and Peng Zou, which establishes how much contact-dependence vs. diffusion is responsible for biotinylation — tl;dr TurboID has a much smaller effective radius! www.nature.com/articles/s41... 1/2
Spatial barcoding reveals reaction radii and contact-dependent mechanism of proximity labeling - Nature Chemical Biology
A DNA nanoruler platform enables precise measurement of the labeling radii of TurboID and APEX2, uncovering their contact-dependent mechanisms. These findings redefine the enzymology of proximity labe...

Happy #FluorescenceFriday! Temporal projection of a time-lapse recording decided to become abstract art. Pretty splashy, isn't it? PalmBow-labeled commissural axons in culture. #neuroscience #microscopy #livecellimaging 🧪
Neat trick if you polycolonal ab's suck. Incubate them with fixed cells with a KO of your protein of interest, then spin. Protocol here: www.med.upenn.edu/markslab/ass... I was amazed how well it worked on first try (I'm sure that I can completely eliminate unspecific bands) #WesternBlot #cellsky
Very happy to share that my latest submission to the #NikonSmallWorld in Motion competition received an Honorable Mention this year! 🥳 #neuroscience 🧪
Huge congratulations to @axonsalex.bsky.social for his Honorable mention a the #NikonSmallWorld in Motion competition! His movie shows axonal growth of chicken dorsal root ganglia neurons using our SPY555-FastAct probe to stain actin. www.nikonsmallworld.com/galleries/20... #FluorescenceFriday
Check it out at PNAS today. Thank you Alexandre for the collaboration!! PK Mem probes offer long time-lapse imaging of cell membrane dynamics. Made possible by Jing Ling, Yitong Liu, and all the collaborators! Available @spirochrome.com
Thrilled to collaborate with @zhixingchen2.bsky.social Lab @pku1898.bsky.social ! We tested new PK Mem dyes—gentle, photostable tools to label the plasma membrane in live neurons, making it easier to track growth cone motility & axonal transport. #liveimaging #neuroscience tiny.cc/3qhp001

Hot off the press: The excellent Maheva Andriatsilavo and colleagues show how a temporal sequence of stochastic molecular mechanisms allow the emergence of stereotyped individualised neuronal circuits. Just published in Nature Neuroscience: rdcu.be/ek010

These ones are growing in a dish, but in a developing embryo or brain, each growth cone would be sniffing its way through a complex molecular terrain, often over long distances to find its correct synaptic partners.
it is a filopodial (thin protusions) party in there! 🕺🧐 🧪 #liveimaging
This time-lapse captures 17 hours of axonal growth from a chicken dorsal root ganglion explant, visualized through the actin cytoskeleton using live confocal imaging. I just submitted this video to the Nikon Small World in Motion competition. Today is the last day to upload yours! 😉 🧪

2024 #FluorescenceFriday retrospective: It was a great year in science for microglia, the brain's immune cells! The brain is much more than neurons; these little cells are essential, particularly in inflammatory processes. Check them interacting below! Markers: neurons (🔵 MAP2), microglia (🟡 IBA1)
How proximal is proximity labeling❓ Using DNA nanorulers we found that TurboID/BioID are largely contact labeling, while APEX2 also bears a major contact labeling pathway in parallel with diffusive labeling. www.biorxiv.org/content/10.1... Special thanks to @aliceyting.bsky.social for discussions
I am super excited to share our article addressing a longstanding question in neurobiology: how Ca²⁺ signaling cascades initiated at dendritic spines operate at a distance. Check it out here: authors.elsevier.com/sd/article/S... @cp-cell.bsky.social.
For this #FluorescenceFriday, let's watch an axonal growth cone (for a change) displaying its most beautiful filopodia (this time)! 🤩 They are so dynamic! 1 image taken every 500 ms for 5 min played at 15 fps. 🧪
I used VVD Viewer for this #FluorescenceFriday video: 141 volumes from the #HHMIJanelia FlyLight Split-GAL4 #Drosophila driver collection. The animation comes from a high-level description processed by neuVid. Such a rapid tour of the data has questionable scientific value, but it's fun to watch!
Lynn Landmesser was a giant in the field of developmental neurobiology. Sad to hear of her passing. #DevBio #neuroscience 🧪
I am saddened by the passing of Lynn Landmesser, a pioneer in neural development and cornerstone of our department. When I arrived, she was still teaching students to fabricate electrodes and dazzling junior faculty with stories about the history of the field. She will be missed.
This axonal growth cone from a culture of spinal neurons decided to display its most beautiful lamellipodia for this #FluorescenceFriday! 🤩 #neuroscience #liveimaging 🧪 1 image taken every second for 10 min, played at 15 images/s
