One of biggest mysteries in biology: how did complex eukaryotic cells evolve from simple microbes? ~1.8 billion years ago, an archaeal cell likely merged with a bacterium to form the first eukaryotic cell, but can we ever find direct evidence of this transformative event? 🦠 🚶‍♂️

For the record, this is my comment to the journalist on Colossal’s dire wolf.

Regarding Colossal's claims on ‘bringing back’ dire wolves: They absolutely and unequivocally have not “brought back” the dire wolf. They have genetically modified grey wolves to have white fur & to reach a greater adult size. Colossal have told us that they made 20 gene edits across 14 genes. Although I do not know which genes were targeted, I do know that twenty gene-edits does not a resurrected species make — there is a far greater genetic gulf between grey wolves and dire wolves than this small number of changes could ever hope to bridge. Khaleesi, Romulus and Remus are not dire wolves by any recognised definition of a species — the pups have not been shown to be ‘phenotypically’ a dire wolf (personal opinion that they ‘look like’ dire wolves is not sufficient evidence); they are not genetically a dire wolf; they are not, evolutionarily speaking, a dire wolf. Unless they can be shown to be morphologically, ecologically, and genetically distinct from grey wolves, then they are grey wolves (Canis lupus). If it should ever occur that such genetically modified wolves can form a population that remains morphologically and genetically distinct from grey wolves over multiple generations, then they should be designated a new species, with a new name. And this speaks to one of the fundamental problems with de-extinction. Even if your goal is just restore ecologically important ’traits’ to an ecosystem - for example by tinkering with an existing species so that they play a different role in the ecosystem. To do this, you need to know what trait cause which ecological effect. We are so far from understanding this complexity. There is, therefore, a logic in saying: “Oh well, we don’t fully understand it, but we do know that this ancient ecosystem was ecologically balanced, and so if we recreate it faithfully then it ought to work.” The minute you deviate from the “recreate faithfully” idea, to “approximate” then you come back to the hard question of what species or traits actually matter. A dire wolf was not just a bigger, whiter wolf. The traits that Colossal have tinkered with are unlikely to result in wolves that can fill the dire-wolf shaped gap in American ecosystems. For example, it is unlikely that dire wolves had white fur. It remains to be seen how the gene edits relating to body size translate into adult body size and shape. Dire wolf behaviour and its role in Ice Age America’s ecosystems is not fully understood. Dire wolves coexisted there with other top predators such as extinct sabre-toothed cats & America Lions, and also lived alongside grey wolves & coyotes — without inter-breeding with them. There was something about its biology that kept it separate from these other species — biologically and ecologically. I doubt it was just body size, as dire wolves and grey wolf body size ranges overlap. There are tantalising hints from the penis bone that maybe mating behaviour (which can affect group structure) was different. And the hyoid bone suggest their calls — so key for species recognition — differed from grey wolves too. But the fossil record can’t tell us everything — and neither can even the most high-coverage ancient genome. It’s not de-extinction, it’s a synthetic biology experiment. First they are testing whether or not the genes selected can produce the desired phenotype (so far, it seems, Colossal have played it safe and chosen well-characterised genes from mice and dogs to bring about the desired phenotypes in their pups and mice; to me this does not reach the bar of having restored extinct ’traits’ e.g. the woolliest of the wooly mice appear to have phenotypes caused by known, modern, mice knockout genes). Down the line it will be an ecological experiment to find out what impact this streamlined phenotype of chosen traits (white coat, larger size in the case of these pups) has on an ecosystem (if any). So no, they are not dire wolves, and it is not de-extinction. Best wishes, Tori
